MOTS-c activates the AMP-activated protein kinase (AMPK) pathway and inhibits the folate cycle, leading to altered de novo purine synthesis . By modulating AMPK, MOTS-c influences cellular energy sensing, glucose uptake, and fatty acid oxidation. In preclinical models, MOTS-c has been shown to reduce pro-inflammatory cytokines and increase anti-inflammatory markers through AMPK-dependent mechanisms. The peptide also affects the MAP kinase/c-Fos pathway, suggesting broad signaling effects beyond simple metabolic regulation.
