TB-500 functions as a G-actin sequestering protein. It binds monomeric globular actin (G-actin) and maintains a substantial pool of actin in an unpolymerized state, available for rapid filament assembly when cells receive migratory signals . This actin regulation is upstream of many cellular processes: wound healing requires fibroblast and keratinocyte migration; angiogenesis depends on endothelial cell motility; immune responses rely on leukocyte trafficking. By controlling actin availability, TB-500 enables the cytoskeletal dynamics that power cell movement. In research models, this mechanism has been studied in dermal wound assays, cardiac injury models, and tendon repair protocols.
