KPV modulates inflammatory signaling through NF-κB pathway inhibition. In macrophage cultures, KPV has been shown to downregulate pro-inflammatory cytokines including IL-1β, IL-6, and TNF-α. The peptide enters cells via the PepT1 transporter and inhibits IκBα phosphorylation directly, preventing NF-κB nuclear translocation. Because KPV lacks melanocortin receptor activity, researchers can study pure anti-inflammatory effects without the confounding receptor activation seen with full-length α-MSH. This receptor-independent mechanism makes KPV unique among anti-inflammatory research peptides.
