Semax activates the transcription of neurotrophins and their receptors in the central nervous system. In rat models of permanent middle cerebral artery occlusion (pMCAO), Semax enhanced Bdnf, TrkA, and TrkC mRNA expression within 3 hours of administration, and upregulated Ntf3 and Ngf transcription at 24 hours . Unlike general neurotrophic stimulants, Semax appears to selectively affect neurotrophin expression in ischemic cortex while having minimal unspecific effects on intact tissue. This selective transcriptional activation suggests a mechanism tied to hypoxia-induced signaling pathways rather than blanket neural stimulation.
